We develop assays for flow cytometry. Typically, the assays include antibodies to define cell type; antibodies that are target epitopes; and dyes (live/dead, cell cycle). A typical development cycle includes:
- Selection of antibodies.
- Testing antibodies for binding and specificity.
- Selection of positive and negative cell controls.
- Identification of appropriate bead-based performance and calibration.
- Optimization of fixation and permeabilization.
- Optimization of staining.
- Sample handling and shipping evaluations.
- Small test with typical samples.
- Write SOP.
We optimize and reduce to an SOP assays from vendors, laboratories, and/or the literature.
- Obtain current copy of existing protocol.
- Identify uncertain or conflicting instructions.
- Determine the fit of existing protocol to current study.
- Resolve uncertainties.
- Identify and obtain reagents.
- Determine sample format (e.g., fresh, cryo-preserveed, fixed).
- Formulate end points and performance criteria.
- Perform provisional protocol.
- Modify protocol as necessary.
- Identify and test performance and calibrator beads and cellular controls.
- Perform updated provisional protocol.
- Write up and log SOP.
We will process samples for studies using new or existing protocols that we have reduced to an SOP. The steps involved are:
- Develop or adopt an assay.
- Process the samples. Batch mode preferred.
- Perform measurements and analyze data.
